Scientists at the University of Liverpool are leading a 1.65 million project to produce and test the first nanomedicines for treating HIV/AIDS.

The research project, funded by the Engineering and Physical Sciences Research Council (EPSRC), aims to produce cheaper, more effective medicines which have fewer side effects and are easier to give to newborns and children.

The new therapy options were generated by modifying existing HIV treatments, called antiretrovirals (ARVs). The University has recently produced ARV drug particles at the nanoscale which potentially reduce the toxicity and variability in the response different patients have to therapies. Drug nanoparticles have been shown to allow smaller doses in other disease areas which opens up possibilities to reduce drug side-effects and the risk of drug resistance. Nanoscale objects are less than one micron in size a human hair is approximately 80 microns in diameter.

Professor Steve Rannard, from the University's Department of Chemistry, said: "Nanomedicines are being used daily to treat a range of conditions around the world. There are, however, no current nanoparticle HIV therapies that are providing this kind of patient benefit. This project is the first step towards taking the nanomedicine options that we have developed out of our labs and into the clinic, representing a significant milestone in the development of new HIV treatments.

"If we can demonstrate real potential from our planned clinical work with healthy volunteers at the Royal Liverpool University Hospital, then our collaboration partner, IOTA NanoSolutions, will take forward the further development and clinical validation of the ARV drug particles in HIV patients. We also aim to test new formulations for children in developing countries, offering HIV patients around the world the prospect of safer, more effective treatments."

Professor Andrew Owen, from the University's Department of Molecular and Clinical Pharmacology, added: "We have integrated an assessment of pharmacology and safety early in the research and this has allowed us to rapidly progress lead options for clinical trials. The work has been conducted with the Medical Research Council (MRC) Centre for Drug Safety Science also based at the University."

"Our data so far looks really exciting, offering the potential to reduce the doses required to control the HIV virus. This work builds on initiatives by Mdecins Sans Frontires and other groups to seek ways to improve ARV therapy and could have real benefits for the safety of ARVs globally. Importantly we also hope to reduce the costs of therapy for resource-limited countries where the burden of disease is highest."

HIV continues to increase in prevalence, with 34 million people currently infected worldwide. The new HIV therapies offer particular hope for treating children with HIV which affects 3.4 million children under the age of 15 years in Sub Saharan Africa. About 90% of infected infants acquire the virus through mother-to-child transmission. Without treatment one third of children die within their first year of life.

There are currently very limited child-appropriate HIV drugs available and existing treatments carry a range of risks for the infant including under or over dosing. The new HIV nanomedicines from the Liverpool team disperse into water, which will make them easier to administer, particularly to newborn babies.

The project will manufacture the ARV nanomedicines using commercially relevant techniques under clinical grade manufacturing conditions. IOTA NanoSolutions was created to further develop and exploit technology originally developed at the University of Liverpool. The company operates a novel nanoparticle synthesis technology, ContraSol and is working with major global pharmaceutical companies. The ARV programme represents a further extension to the ongoing collaboration between the University of Liverpool and IOTA NanoSolutions.

The project aims to deliver highly valuable data within three years and provide a platform for continual development and testing during that time.

(AP) ? A 15-year-old sophomore at a suburban Baltimore high school who made references to murder-suicide on Facebook has been charged as an adult in the shooting of a classmate on the first day of school, authorities said Tuesday.

Robert Wayne Gladden Jr. was being held without bail on charges of attempted first-degree murder and first-degree assault, Baltimore County police said. A preliminary hearing was scheduled for Sept. 7. The state's attorney's office did not know if he had a lawyer.

Gladden's last status update on his Facebook page, posted the morning of the shooting, read: "First day of school, last day of my life. ... f--- the world."

His father told The Associated Press that his son had been bullied. Baltimore County Police Chief James Johnson said at a news conference Tuesday that he was aware of the reported bullying, but he said Gladden has not indicated in conversations with detectives that bullying was a motive for the shooting. He did not disclose other possible motives.

Gladden continues to cooperate with investigators and was undergoing a mental health evaluation, Johnson said.

Gladden rode to school on the bus Monday morning with a bag containing a disassembled shotgun, 21 rounds of ammunition and a bottle of vodka, Johnson said.

When he arrived at school, Gladden went to his first two classes, Johnson said. On the way to the cafeteria, he stashed the bag with the shotgun in a restroom, the chief said. A short time later, he returned to the restroom and assembled the gun, which he then hid beneath his clothes, Johnson said.

Upon entering the cafeteria, he pulled out the gun and fired a shot toward a lunch table, according to charging documents. A 17-year-old classmate, Daniel Borowy, was struck in the back.

Teachers and school staff then rushed toward the teen, and in the ensuing struggle, he fired another shot that hit the ceiling, investigators said in the documents. The staffers were able to get the gun away from him and he was arrested by a school resource officer.

Witnesses credited guidance counselor Jesse Wasmer with getting the gun away from Gladden.

Gladden sipped from the vodka bottle before the shooting but did not drink enough to become intoxicated, Johnson said.

The teen obtained the shotgun from his father's house, Johnson said. The weapon was manufactured before 1968 and was of legal length, and police were trying to determine whether it was properly registered, he said.

On his Facebook page, Gladden referred to mass murderer Charles Manson and gave himself the nickname "SuicidalSmile." The page, identified by classmates as belonging to the suspected shooter, was just launched in July, and the three photos of Gladden all show his face hidden behind long, dark hair. He describes himself as a "metalhead" and a fan of musicians Marilyn Manson and Slipknot.

The suspect's father, Robert W. Gladden, told the AP Monday evening that his son was the shooter and indicated his son had been bullied. He gave no further details.

A woman who was also at the home in Middle River and said she was related to the elder Gladden gave the following statement on the family's behalf: "We are horrified. We did not see this coming and our thoughts and prayers are with the victim and the victim's family."

Borowy remained in critical condition Tuesday at Maryland Shock Trauma Center, a hospital spokeswoman said. His family issued a statement asking well-wishers to keep him in their thoughts and prayers and asking for privacy.

Police also executed a search warrant at the Kingsville home of Gladden's mother and stepfather and arrested the stepfather ? Andrew Piper, 43 ? on illegal gun and drug possession charges, police said. The charges against Piper had nothing to do with the school shooting, Johnson said, but he noted that Gladden lived part-time at that address.

Classes resumed Tuesday at the school amid a low-key police presence. About 150 students turned out for a prayer vigil organized by local churches on the school grounds. Some students wore T-shirts and bracelets reading "Pray for Daniel" and "Team Wasmer" in reference to the victim and the guidance counselor.

Shane Boyer, 44, who was dropping off his 13-year-old daughter Corinne, said the vigil helped calm students still dealing with the shooting. Boyer said he knew immediately that his daughter was safe after the shooting because he received text messages from her. One read, "Someone just shot a gun in lunch," followed by another that said, "I ran out I am do (sic) scared there's helicopters + cops + we are all outside."

50 buildings destroyed in Northern Calif. fire

MANTON, Calif. (AP) ? Dozens of buildings, many of them likely homes, have been destroyed in recent days a fire burning outside the Northern California community of Manton, fire officials said Tuesday night.

Fire crews assessing the rural area determined Tuesday that 50 buildings had been destroyed, state fire spokesman Daniel Berlant said. The count included buildings burned since the fire began, but officials did not say when the structures were destroyed.

Officials didn't have an accurate count yet of how many of the structures were homes, but Berlant noted the buildings were spread across a vast rural area of mostly residential homes.

The blaze, which was sparked by lightning on Saturday has consumed more than 33 square miles and continues to threaten hundreds of homes.

Nearly 1,900 firefighters were battling the fire in rugged, densely forested terrain as it threatened 3,500 homes in the remote towns of Shingletown, Manton and Viola, about 170 miles north of Sacramento.

As a the wildfire raged near Lynn Rodgers' home of less than a year, the evacuated resident said Tuesday she remained optimistic ? in spite of her growing frustration and fear.

"Yeah, but what can you do? Everything is in God's hands ? and the firefighters," said Rodgers, who lives in Shingletown.

The fast-moving fire is one of many burning across the West, where dry lightning has sparked up grass, brush and timber, bringing an early start to the fire season.

Gov. Jerry Brown announced Tuesday that National Guard troops will be assisting with the firefighting efforts. The news comes a day after the Federal Emergency Management Agency said it is offering federal funds to help fight the blaze.

The fire forced the closure of Highway 44 and other roads, and prompted the declaration of an emergency in Shasta County.

Elsewhere in California, a massive wildfire in Plumas National Forest continued to expand, helped by gusty winds. The blaze, about 120 miles north of Sacramento, has consumed nearly 98 square miles since it started at the end of July and threatens about 900 homes.

In Mendocino County, the sheriff's office issued a mandatory evacuation for residents in Covelo due to a wildfire that has burned more than 15 square miles of thick timber and rugged terrain. One outbuilding has been destroyed and 45 homes were threatened by the blaze, officials said. The fire was sparked by lightning Saturday in a remote area, making it difficult for fire crews to access.

In Washington state, the National Weather Service warned about extreme fire danger in the eastern part of the state as forecasts called for thunderstorms with lightning.

Fire crews to the north were still hoping to fully contain a week-old wildfire that has destroyed 51 homes and 26 outbuildings, and damaged at least six other homes, fire information officer Mark Morrow said.

The fire, about 75 miles east of Seattle, has caused an estimated $8.3 million in property damage.

Lightning over the weekend also sparked smaller fires in Colorado, Idaho and Utah.

The code determines the recognition of RNA molecules by a superfamily of RNA-binding proteins called pentatricopeptide repeat (PPR) proteins.

When a gene is switched on, it is copied into RNA. This RNA is then used to make proteins that are required by the organism for all of its vital functions. If a gene is defective, its RNA copy and the proteins made from this will also be defective. This forms the basis of many terrible genetic disorders in humans.

RNA-binding PPR proteins could revolutionise the way we treat disease. Their secret is their versatility - they can find and bind a specific RNA molecule, and have the capacity to correct it if it is defective, or destroy it if it is detrimental. They can also help ramp up production of proteins required for growth and development.

The new paper in PLOS Genetics describes for the first time how PPR proteins recognise their RNA targets via an easy-to-understand code. This mechanism mimics the simplicity and predictability of the pairing between DNA strands described by Watson and Crick 60 years ago, but at a protein/RNA interface.

This exceptional breakthrough comes from an international, interdisciplinary research team including UWA researchers Professor Ian Small and Aaron Yap from the ARC Centre for Excellence in Plant Energy Biology and Professor Charlie Bond and Yee Seng Chong from UWA's School of Chemistry and Biochemistry, along with Professor Alice Barkan's team at the University of Oregon. This research was publicly funded by the ARC and the WA State Government in Australia and the NSF in the USA.

"Many PPR proteins are vitally important, but we don't know what they do. Now we've cracked the code, we can find out," said ARC Plant Energy Biology Director Ian Small.

"What's more, we can now design our own synthetic proteins to target any RNA sequence we choose - this should allow us to control the expression of genes in new ways that just weren't available before. The potential is really exciting."

"This discovery was made in plants but is applicable across many species as PPR proteins are found in humans and animals too," says Professor Bond.

The hands appear to be the first physical evidence of a practice attested to in ancient Egyptian writing and art, in which a soldier would present the cut-off right hand of an enemy in exchange for gold, Bietak explains in the most recent edition of the periodical Egyptian Archaeology.

"Our evidence is the earliest evidence and the only physical evidence at all," Bietak said. "Each pit represents a ceremony."

Scientists are not certain who started this gruesome tradition. No records of the practice have been found in the Hyksos' likely homeland of northern Canaan, Bietak said, so it could have been an Egyptian tradition they picked up, or vice versa, or it could have originated from somewhere else.?

Bietak pointed out that, while this find is the earliest evidence of this practice, the grisly treatment of prisoners in ancient Egypt was nothing new. The Narmer Palette, an object dating to the time of the unification of ancient Egypt about 5,000 years ago, shows decapitated prisoners and a pharaoh about to smash the head of a kneeling man.?
